Man OP has zero rizz, has a bunch of unlisted criteria and seems just like the type of guy you can't have a conversation with..... You've been given several answers, just take them.

Your question has been answered several times I feel, where people gave you /their/ hardest missions in RDR2( y'know,, the answer to the question?), BUT only after they answered did you change the question to be specifically main quest missions, when most people have out of the box answers like, completeionist and turning the game off. Your responses to these answers are just very dry and frankly hard to think of any response for, if you wanted a specific answer you should have asked a more specific question. Not to mention the way you're responding just in general seems very short and rude, but I haven't played the game so I can't give an answer, I was just annoyed by what I saw in the comments, sorry for being a dick.
I respect the quick response though, that's always nice.

I didn't find any particularly hard, but I did fail several due to buggy controls.
My horse would just start randomly spinning in circles during chase/escort missions and would stop, so I would have to fail and reload the mission. >! It happened so much that I cheered when the horse died.!<
Controls
I think the reason why everyone is giving similar answers and why possibly you can’t think of one yourself is that RDR2 is designed as a cinematic, narrative experience where no single mission is designed to be much harder than the other (except for the collectible based stuff naturally). They’re all kinda tuned to be “middling” difficulty so that the player doesn’t get bored but also isn’t seriously impeded from continuing the main story.
It’s also a mega long game that the majority of us haven’t likely touched in great depth since it came out 4-5 years ago. The dedicated subreddit might know more as those folks will be more invested into the game.
If you still want an answer, the attack on the O’Driscolls (I think that’s what they’re called) while you’re still in the snowy area at the start. Unfortunately for your purposes it’s for no other reason than being a mission with a lot of enemies while you’re still getting used to this amazing game’s, awful controls .

That one mission that’s in a blizzard and it’s more of a prologue sorta mission but it was so hard
Fighting the arbitrary rules that R* puts in place for missions. I got really tired of seeing “Mission Failed” because I looted bodies like the game taught me to do, or simply didn’t approach the missions exactly how they wanted me to do so. That was incredibly draining and why I don’t think the game is as great as people continually claim it to be.
Sure, it’s a great cinematic experience, but it’s frustrating to see a open world turn into an on the rails ride.
I agree, the first Red Dead Redemption was so rich in it's gameplay then they turned the second into a theme park ride.
Side mission? Probably finding all the wind chimes in the Wisdom of the Elders.
Main quest? I remember John Martin’s final charge up the mountain being difficult

You either go somewhere and talk/escort some npc’s. Maybe do a unique mini game, or you go somewhere and shoot people.
If it’s a robbery missions like a bank or train, it’s much more scripted and it’s only difficult if you don’t follow the intended script
